A master’s thesis at the Faculty of Medicine, Karbala University, investigated the role of heat shock protein (70) and vitamin D3-binding protein and its relationship to vitamin D3 receptor genes in obese women with type 2 diabetes.
Where the thesis presented by the researcher Baraa Abdulkarim Matar Hussain included “The heat shock protein-70 is a highly conserved family of stress-responsive proteins. It was revealed that HSP-70 plays a function in modulating conditions associated with chronic diseases such as diabetes.” As for heat shock proteins ( HSP) have molecular weights ranging from 15 to 110 kDa and are widespread and well-known proteins. Heat shock proteins act as chaperone proteins and can assist in the remodeling of denatured proteins. Heat shock proteins play important roles in providing defense mechanisms against diseases, particularly diabetes.
The study also came with the aim of the study, as it includes examining the levels of VDBP protein, vitamin D3, the association among Iraqi women with type 2 diabetes, and comparing them with the control group. Molecular studies include identification of single nucleotide polymorphisms of the vitamin D3 gene in obese women with type 2 diabetes by genetic sequencing. Serum levels of insulin, glucose, HbA1C%, and heat shock protein-70 were determined. 50 associations between different phenotypes and genotype outcomes were also studied. This case-control study was conducted on 50 patients with type 2 diabetes who attended Al-Hussein Teaching Hospital and Al-Hassan Center for Diabetes and Endocrinology / Karbala Health Directorate – Iraq and another individual with diabetes, during the period from April 2022 to April. 2023). T2DM groups were divided into two groups, 25 obese and 25 non-obese. The control group was divided into 25 obese and 25 non-obese subjects as healthy-looking groups. 5 ml of blood was aspirated from each patient, 3 ml of blood was centrifuged and serum was used to determine C, HbA1c, RBS S-70 Peptide (25) and insulin. ). The remaining 2 mL of blood was deposited
into an EDTA tube for DNA extraction.
The observed results were that the levels of 70-HSP and VDBP showed a non-significant decrease in all groups of patients with type 2 diabetes compared to the control group, while C-peptide decreased significantly in patients with type 2 diabetes when compared with the control group. In addition, both random glucose and blood HbA1c showed a significant elevation in patients with type 2 diabetes compared to apparently healthy control groups.
